RPGs with semi-randomized stories?
I'm not aware of any RPG doing this(?), but I was always fascinated by the Bladerunner game having a semi-randomized story. For those that have not played it/are unaware, whether certain characters are a human or replicant is randomized and the story adjusts for this.
I know Underrail has a chance of having certain areas available or not, along with some Diablo quests, but this is not the same thing as it's just locking you out of content. I guess the Bladerunner example is more of a story randomizer except the writers have accounted for all the variants.

Lost Dimension comes to mind. It's a JRPG that advertised a feature where you are in a party with eleven other characters five of which will betray the group. The five are randomly selected each time you start a new game and it's up to you to deduce who the traitors are.
Splintered is an RPG with an unlockable roguelike mode. You beat the game several times over the course of the story, and each time you have new classes available, and various elements of the plot are randomized. The map, storyline item locations, and shop vendors all change from play to play. It can result in wildly varying playthrough difficulty. I enjoyed playing through the full story but probably won't ever bother with the roguelike mode.
It's not quite what you asked for but it's the closest I could think of.
Silent Storm has randomly generated plot based on you going through missions that give clues for further missions. It can place same clues on various maps and in varying circumstances. E.g. in one playthrough you may capture a suspect officer in one of the first missions, in other playthrough he may evade you until almost the end of the game to spawn on a completely different map.
Plot-relevant missions are premade, though. Only minor things differ.
